Se ha especulado mucho sobre el móvil de este crimen, que aún hoy no está demasiado claro. En principio se dijo que la elección de la casa había sido casual. Se sabe que Manson ya había estado en aquella mansión por lo menos en dos ocasiones.
Se dice que el posible móvil fue el rodaje de la película de Roman Polanski (http://es.wikipedia.org/wiki/Roman_Polanski) Rosemary's Baby (http://es.wikipedia.org/wiki/Rosemary%27s_Baby_%28pel%C3%ADcula%29). El director había sufrido amenazas a causa del rodaje por grupos esotéricos de la época, ya que trataba un tema polémico como era la práctica del satanismo entre las personas de la élite americana y el advenimiento de un hijo del diablo al igual que Dios mandara a Jesús a la Tierra.
En las paredes de la habitación los asesinos escribieron con sangre de sus víctimas como "Pig" (Cerdo) y "Helter Skelter".
Actualmente se descubrió que el móvil de los asesinatos tenían que ver una venganza personal contra el peluquero de Sharon Tate, Jay Sebring, quién habría pagado a un par de mujeres (supuestas seguridoras de Manson) para que se dejaran "humillar sexualmente" por Sebring. En venganza, los miembros de La Familia, se dirigieron al lugar dónde se encontraba el peluquero y, confundiendo a Tate como una amante, decidieron darle muerte ante él.
